Alciporin, a pore-forming protein as complementary defense mechanism in Millepora alcicornis

چکیده

Millepora alcicornis (Cnidaria: Hydrozoa), known as fire coral, is a tropical species settled in marine ecosystems of the Canary Islands last years. This hydrocoral biosynthesizes toxins involved chemical defense and prey capture mechanisms. Toxicological studies have shown that venom contained nematocysts mainly composed thermolabile proteins display hemolytic activity, causing skin irritation burn-like lesions upon contact. As continuation previous study, chromatographic fractionation aqueous extracts M. has confirmed coexistence different nature responsible for effects red blood cells (RBCs) through two Aside from already described phospholipase A2 (PLA2) this work presence alciporin, pore-forming protein (PFP), been established first time . The sequence analysis revealed alciporin fit an actinoporin with high homology to stichotoxins. were analyzed sphingomyelin was identified its biological target. Also, evolution damage produced at nanoscale studied using atomic force microscopy (AFM).

Engineering a pH responsive pore forming protein

Listeriolysin O (LLO) is a cytolysin capable of forming pores in cholesterol-rich lipid membranes of host cells. It is conveniently suited for engineering a pH-governed responsiveness, due to a pH sensor identified in its structure that was shown before to affect its stability. Here we introduced a new level of control of its hemolytic activity by making a variant with hemolytic activity that w...

Pore Formation Mechanism of Staphylococcal Pore- forming Toxin

Y. Tanaka and M. Yao (Hokkaido Univ.) Pathogenic bacteria express pore-forming toxins (PFTs) to attack host cells. PFTs are expressed as soluble monomeric proteins, which assemble to prepore oligomer on the target cells. After forming prepore, conformational change occurs, and then the pore is formed. Although the crystal structures of monomer and pore have been determined, the detailed mechani...

Diarylthiophenes as inhibitors of the pore-forming protein perforin

Evolution from a furan-containing high-throughput screen (HTS) hit (1) resulted in isobenzofuran-1(3H)-one (2) as a potent inhibitor of the function of both isolated perforin protein and perforin delivered in situ by intact KHYG-1 NK cells. In the current study, structure-activity relationship (SAR) development towards a novel series of diarylthiophene analogues has continued through the use of...

Protein washdown as a defense mechanism against myocardial edema.

Myocardial edema occurs in many pathological conditions. We hypothesized that protein washdown at the myocardial microvascular exchange barrier would change the distribution of interstitial proteins from large to small molecules and diminish the effect of washdown on the colloid osmotic pressure (COP) of interstitial fluid and lymph. Dogs were instrumented with coronary sinus balloon-tipped cat...

Response of Millepora alcicornis (Milleporina: Milleporidae) to two bleaching events at Puerto Morelos reef, Mexican Caribbean.

Two naturally occurring colonies of Millepora alcicornis were monitored during 1997 and 1998, both years in which this species bleached in the Mexican Caribbean. One colony (HL) was naturally exposed to a high light environment and another nearby colony (LL) was exposed to 5.9 times lower light levels due to shadowing by a pier. For 10 days in August 1997, seawater temperatures in the surroundi...
